A few have asked for a more 30K Ft view of this. If you recall process 1 vs process 2 bait and switch described by @RetsefL and @joshg99… The PCR step in process 1 erases all methyl groups on DNA. Switch to process 2 and you reintroduce not just SV40 and the whole plasmid backbone but now all your residual DNA looks like a foreign invader to your cells as it’s methylated in a manner not seen in humans. https://twitter.com/kevin_mckernan/status/1974821167857877416
